A high-resolution transcript profile across the wood-forming meristem of poplar identifies potential regulators of cambial stem cell identity
-
J. Schrader, J. Nilsson, E. Mellerowicz, A. Berglund, P. Nilsson, M. Hertzberg, and G. Sandberg A high-resolution transcript profile across the wood-forming meristem of poplar identifies potential regulators of cambial stem cell identity Plant Cell 16 2004 2278 2292 This paper is exemplary in that it applies cutting edge technology to unravel the biology of a poorly understood tissue typical for perennial plants. By cryosectioning of the vascular cambium [27], single-cell layers were obtained from which RNA was prepared for microarray hybridizations. Transcript profiling reveals different transcriptomes for the six anatomically similar cambial cell layers.

The work is special in that the samples are derived from outdoor-grown trees and thus reflect both developmental and environmental cues. Such studies are essential to understand the full biology of trees in their natural environment.

Coordinated genetic regulation of growth and lignin revealed by quantitative trait locus analysis of cDNA microarray data in an interspecific backcross of eucalyptus

Sederoff Coordinated genetic regulation of growth and lignin revealed by quantitative trait locus analysis of cDNA microarray data in an interspecific backcross of eucalyptus Plant Physiol 135 2004 2368 2378 The first paper describing a genetical genomics approach in forest trees to identify candidate genes for growth. The study was carried out in an interspecific Eucalyptus backcross population, in which QTLs for growth co-localized with QTLs for mRNA abundance of lignin biosynthesis genes and associated methylation pathways. SAMS turned out to be an interesting candidate gene for further functional analysis.

Field and pulping performances of transgenic trees with altered lignification

G. Pilate, E. Guiney, K. Holt, M. Petit-Conil, C. Lapierre, J.-C. Leplé, B. Pollet, I. Mila, E.A. Webster, and H.G. Marstorp Field and pulping performances of transgenic trees with altered lignification Nat Biotechnol 20 2002 607 612 This paper reports data from four-year old field trial experiments, carried out in two countries, with transgenic poplar modified in lignin biosynthesis. The data show that wood from poplars downregulated for CAD requires fewer chemicals to extract a given amount of lignin from the pulp, which could have both economic and environmental benefits. Importantly, no adverse phenotypes on plant growth and health were noticed during the four-year period in the field.
