A refined structure of the complex suggests the mechanism of downstream DNA unwinding. A non-reactive NTP analogue was located in a pre-insertion site, which is close to the insertion site. The elongation factor TFIIS is shown to reposition RNA in the active center. This is the first structural indication that alternative conformations of the EC exist that reflect different functional states of the enzyme.

The authors suggest that substrate selection occurs before isomerization to a catalytically competent conformation.
