The first structure of a type III secretion chaperone, SicP, bound to its binding region on the effector SptP of Salmonella enterica, is presented. The results are enlightening, even though it is not the structure of the entire SptP protein (543 residues), but that of SptP35-139, which has been analysed.

Nat Struct Biol 2001, 8:1031-1036. This paper provides a comparison of the crystal structures of two type III secretion chaperones from Salmonella enterica and enteropathogenic Escherichia coli (EPEC), and reveals striking similarities. The authors also show that the physical properties of effectors are similar in the presence and in the absence of their respective chaperones, indicating that binding of the chaperone to the substrate does not have a global antifolding effect on the entire substrate.

The structure of the Yersinia spp. chaperone SycE bound to the chaperone-binding region of YopE is compared to that of SycE alone (see also Evdokimov et al. [2002] [9]). The authors also provide evidence that the binding of the chaperone affects only the conformation of its binding region on the effector (as in Luo et al. [2001] [8••]). The discussion on the participation of the chaperone in the secretion signal and the temporal order of secretion is interesting but highly speculative.
