The IcrQ mutants were constructed with a double polymerase chain reaction (PCR) strategy described in detail elsewhere (12). In short, the IcrQ gene was exchanged with the streptomycin-spectinomycin (Sp/Sm) resistance gene which was excised from mini-Tn5-Sm/Sp (26), The IcrQ deletion construct was introduced into the virulence plasmids pIB102 and pIB68 by conjugation and allelic exchange with the suicide vector pDM4 (27). The resulting virulence plasmids are denoted pIB26 (IcrQ) and pIB6826 (yscS/IcrQ), respectively. The IcrQ-IuxAB operon fusion was constructed with the same strategy as that described for the yopE-IuxAB fusion (19).
