    • Antibody multispecificity mediated by conformational diversity
    • The binding of IgE to FcεRI without the addition of antigen stimulates mast cell proliferation without overt degranulation. This paper shows that the crystal structure of the anti-DNP IgE, which has strong activity for this function, has multiple conformations, suggesting that the Fab of this IgE may bind with low affinity to different cell surface proteins to activate cellular enzymes that result in intracellular signals.

    • •] describe the function of SWAP-70 and its expression in mast cells. This protein is shown to be a guanine-nucleotide exchange factor and is present in immature but not mature mast cells and BMMCs. There is decreased degranulation of SWAP-70-negative BMMCs; however, the lack of SWAP-70 in mature mast cells suggests that SWAP-70 is not essential for degranulation.

    • These studies further dissect the essential role of SLP-76 in FcεRI-induced signaling. The amino-terminal and GADS-binding domains of SLP-76, but not the SH2 domain, are essential for FcεRI-mediated degranulation and IL-6 secretion. However, there are differences between the SLP-76 domains required for degranulation and those required for the release of IL-6.

    • Absence of platelet endothelial cell adhesion molecule-1 (CD31) leads to increased severity of local and systemic IgE-mediated anaphylaxis and modulation of mast cell activation
    • Both in vivo and in vitro allergic reactions are enhanced in PECAM-1-null mice, suggesting that this protein is a negative regulator of FcεRI-mediated reactions.
