In this report the authors, using fluid-shear stress, hyperosmotic shock and anti-PECAM-1-coated magnetic beads, demonstrate PECAM-1 and ERK phosphorylation and PECAM-1/SHP-2 binding and suggest that in addition to its other functions PECAM-1 may function as a mechanotransducer, affecting pivotal signaling cascades.

PECAM-1's interaction with SHP-2 was also found to influence SHP-2's substrate specificity, including tyrosine-phosphorylated β-catenin and FAK, thus affecting endothelial migration.

Tissue-specific distributions of alternatively spliced human PECAM-1 isoforms

These two papers demonstrate the complex, partially species-specific alternative splicing patterns observed in the cytoplasmic domain of PECAM-1 during development and differentiation. Interestingly, the alternatively spliced exons (12-15) correspond to those mediating SHP-2, src family member, β- and γ-catenin and STAT family member interactions.

During the transendothelial migration of leukocytes, this membranous network provides the migrating leukocytes with sufficient PECAM-1/PECAM-1 interaction sites during transmigration and is recycled to this compartment once transmigration is complete.
