    • The membrane-proximal region of the E cadherin cytoplasmic domain prevents dimerization and negatively regulates adhesion activity
    • Using the leukemia cell line K562, these researchers found the juxtamembrane region of E-cadherin negatively regulates adhesive properties of the cell. Deletion of the juxtamembrane region resulted in enhanced lateral dimerization, with calcium-dependent aggregation, but compaction did not occur.

    • Adhesive but not lateral E-cadherin complexes require calcium and catenins for their formation
    • This study shows that the formation of lateral E-cadherin dimers is independent of catenin, calcium and cell-cell contact. Lateral dimers associate between opposing cells in an anti-parallel fashion. This configuration is catenin and calcium dependent and affords significant adhesive strength between cells. Formation of lateral dimers is proposed to precede formation of the adhesive complex.

    • Functional domains of α-catenin required for the strong state of cadherin-based cell adhesion
    • Three distinct functional domains for α-catenin were identified: the vinculin binding domain, the ZO-1 binding domain and the adhesion modulation domain. In 'epithelialized' L cells, the authors found ZO-1 and the adhesion modulation domain confer strong cell adhesion. Although the ZO-1 domain did not support strong cell adhesion itself, they suggested that α-catenin's interaction with the cytoskeleton through the ZO-1 domain is required for strong E-cadherin based cell adhesion.
