The findings demonstrate that resting T cells in the absence of specific antigen stimulation may contribute to the activation of synovial fibroblasts in conditions, in which large numbers of T cells are in direct contact with these cells.

Arthritis Rheum 2001, 44:1555-1567. The data are important because they demonstrate that, unlike in osteoarthritis synovial fibroblasts, the protein kinase Akt may act as a survival signal in RA-SF. Moreover, downstream signaling molecules such as Akt are suggested to be key regulators of the apoptotic response to TNF-α in RA-SF.

In this study, microdissected synovial tissues from RA patients were analyzed for the presence and distribution of p53 mutations. Abundant p53 transition mutations were found in clustered areas together with increased expression of IL-6. These data appear to confirm previous observations on the presence of p53 mutations in RA and link these to the oligoclonal expansion of RA-SF as well as increased cytokine production.
