    • Ali M, Ponchel F, Wilson KE, et al.: Rheumatoid arthritis synovial T cells regulate transcription of several genes associated with antigen-induced anergy. J Clin Invest 2001, 107:519-528. The authors identify transcriptional changes that are associated with T cell anergy in RA synovial T cells. Specifically, they provide a link between the downregulation of calmodulin and impaired T cell responsiveness in RA. This interesting paper supports the concept of T cell anergy in the rheumatoid synovium.

    • Yamamura Y, Gupta R, Morita Y, et al.: Effector function of resting T cells: activation of synovial fibroblasts. J Immunol 2001, 166:2270-2275. This paper demonstrates the upregulation of cytokines and MMPs in RA-SF by resting T cells. The findings demonstrate that resting T cells in the absence of specific antigen stimulation may contribute to the activation of synovial fibroblasts in conditions, in which large numbers of T cells are in direct contact with these cells.

J Immunol 2001, 167:3064-3073. The authors provide strong evidence that SDF-1, as produced by fibroblast-like cells, modulates the behavior of T cells. Consequently, this paper strengthens the concept that RA-SF may contribute significantly to T cell accumulation in the rheumatoid synovium.

The data are not related directly to arthritis, but the demonstration that Bcl-2 regulates a caspase activation program independently of the cytochrome c/Apaf-1/caspase-9 complex is of fundamental importance and may affect apoptosis research in RA, where Bcl-2 has been found to be upregulated in a variety of synovial cells.

J Immunol 2002, 169:1760-1767. This paper shows that autocrine stimulation of RA-SF with IL-15 results in the overexpression of antiapoptotic members of the Bcl-family and contributes to the resistance of RA-SF to apoptosis. The data provide another important link between stable activation of RA-SF and the cytokine network.

    • Regulation of tumor necrosis factor alpha-mediated apoptosis of rheumatoid arthritis synovial fibroblasts by the protein kinase Akt
    • Zhang HG, Wang Y, Xie JF, et al.: Regulation of tumor necrosis factor alpha-mediated apoptosis of rheumatoid arthritis synovial fibroblasts by the protein kinase Akt. Arthritis Rheum 2001, 44:1555-1567. The data are important because they demonstrate that, unlike in osteoarthritis synovial fibroblasts, the protein kinase Akt may act as a survival signal in RA-SF. Moreover, downstream signaling molecules such as Akt are suggested to be key regulators of the apoptotic response to TNF-α in RA-SF.

    • Yamanishi Y, Boyle DL, Rosengren S, et al.: Regional analysis of p53 mutations in rheumatoid arthritis synovium. Proc Natl Acad Sci USA 2002, 99:10025-10030. In this study, microdissected synovial tissues from RA patients were analyzed for the presence and distribution of p53 mutations. Abundant p53 transition mutations were found in clustered areas together with increased expression of IL-6. These data appear to confirm previous observations on the presence of p53 mutations in RA and link these to the oligoclonal expansion of RA-SF as well as increased cytokine production.
