    • 13C-labeling. Metab Eng 2001, 3:151-162. This paper provides a timely reminder of potential problems in the use of steady-state labelling strategies for flux determination. The authors show that exchange reactions catalysed by transketolase and transaldolase that have no net stoichiometric effect (and thus are often ignored) influence the labelling pattern of metabolites. Failure to consider these exchange reactions can lead to erroneous estimates of flux throughout the metabolic network, even though the calculated flux values might provide a good fit to the observed labelling pattern.

    • Klamt S, Stelling J: Two approaches for metabolic pathway analysis? Trends Biotechnol 2003, 21:64-69. A considered assessment of two recent conceptual frameworks for pathway analysis. This is an ideal introduction to the development of new ideas on how to visualise metabolism. Klamt and Stelling conclude that metabolic pathways are best examined in terms of 'elementary flux modes' and argue that, under conditions in which analyses based on the identification of 'extreme pathways' are applicable, the two approaches are synonymous.

Planta 2001, 212:499-507. A specific isozyme of plastidic glucose 6-phosphate dehydrogenase is shown to be differentially expressed in different organs throughout the plant. In both roots and leaves, the expression of this isozyme increases transiently in response to nitrate application. The 5′-untranslated region of the gene that encodes the isozyme contains several NIT2-binding motifs, suggesting that the stimulation of isozyme expression may be due to the direct regulation of transcription by nitrate.

Eur J Biochem 2001, 268:2678-2686. The authors describe the purification of chloroplast 6-phosphogluconate dehydrogenase, and the isolation of the corresponding cDNA from spinach. This work allows the direct comparison of the sequences of the plastid and cytosolic forms of this enzyme. Phylogenetic analysis reveals that both the plastid and the cytosolic isozymes from higher plants are more similar to cyanobacterial homologues than to animal or fungal cytosolic enzymes. The authors propose, therefore, that the ancestral gene for higher plant 6-phosphogluconate dehydrogenase was obtained from the'antecedent of plastids through endosymbiosis and gene transfer to the nucleus. They further propose that the cytosolic form of the higher plant enzyme was derived secondarily through gene duplication and replacement of a pre-existing cytosolic homologue.

This work establishes the importance of plastid transketolase, which catalyses a near-equilibrium reaction, as a determinant of photosynthesis and phenylpropanoid metabolism. The explanation for how changes in transketolase expression influence these metabolic pathways has much broader significance: it highlights the sensitivity of metabolic processes to changes in the concentrations of metabolites, including those that are not directly associated with the specific pathways under study.

This activity, which occurs in a wide range of tissues, is likely to allow the exchange of intermediates between the cytosolic and plastidic pools of pentose phosphates, and thus to contribute to the integration of pentose phosphate metabolism within these compartments.

This work provides an excellent example of how knowledge of the genome sequence can be used to inform and direct a comprehensive study of the structure and function of various members of a specific gene family.

It is therefore abolished by inhibitors of glutamine synthase and mimicked by growth on glutamine. Kinetic analysis suggests that a plastidic form of the enzyme is induced.
