Cell. 108:2002;615-627. • The authors perform gel mobility shift experiments using DNA-bending probes, in combination with TBP mutants to define a two-step process for the recognition and bending of DNA by TBP. They show that the N terminus inhibits the DNA-bending step in promoter recognition. A solvent-exposed surface was identified that inhibits the transition to the stable bent DNA complex. The authors provide evidence that this process may be regulated by cellular factors, as TFIIB can convert the unbent TBP-DNA complex into the bent complex.

A TAF octamer was formed by pair-wise expression of the heterodimerization partners in bacteria, followed by in vitro reconstitution of the heterodimers. The physical properties and proper stoichiometry of the subunits were confirmed by multiple biochemical analyses.

Using conformation-specific antibodies that recognize epitopes within the linker within human TFIIB in gel mobility shift assays, the authors show that the binding of these antibodies to IIB within the context of a TBP-TFIIB-DNA complex (TB-DNA) is affected by sequences directly adjacent to the TATA sequences. Conformation-specific antibodies failed to super-shift TB complexes formed on the two promoters containing a strong BRE element (AdML and hIGFII), whereas they were able to recognize TB complexes formed on core promoters lacking these elements. Also, they demonstrate that TFIIB mutants defective for start site utilization adopt a different conformation than wild type IIB.
