This article shows that dietary diacylglycerol (30% diacylglycerol + 13% sucrose) suppressed high fat and a high sucrose diet (30% triacylglycerol + 13% sucrose) induced body fat accumulation in C57BL/6J mice. Circulating leptin and insulin levels were also reduced to the respective control levels (5% triacylglycerol fed). High diacylglycerol feeding suppressed the elevation of leptin mRNA in adipose tissue and upregulated β-oxidation related genes.

A randomised, double-blind, parallel intervention trial with 131 overweight or obese men which shows an ingestion of foods containing diacylglycerol oil during 24 weeks promotes weight loss and body fat reduction and may be useful as an adjunct to diet therapy in the management of obesity.

This was a randomized crossover designed trial with 12 healthy normolipidemic or moderately hypercholesterolemic men which showed hypocholesterolemic activity of mayonnaise containing phytosterols (500 g/day) dissolved in diacylglycerol (10 g/day) superior to that containing phytosterols (500 g/day) dissolved in triacylglycerol (10 g/day). Higher solubilized phytosterol into the diacylglycerol may interfere with dietary cholesterol in the process of micene formation in the intestinal lumen.

The authors also found that diacylglycerol feeding stimulates β-oxidation and lipid metabolism-related gene expression and uncoupling protein-2 messenger RNA in the small intestine of C57BL/6J mice within the first 10 days of feeding.
