    • Causal relationship between the loss of RUNX3 expression and gastric cancer
    • -/- mice show hyperplasia in the glandular stomach epithelial cells, which appears to be due to insensitivity of the cells to the growth-inhibitory activity and the apoptosis-inducing activity of TGF-β. As RUNX proteins are integral components of the TGF-β signaling cascade, the authors suggest that RUNX3 is a new addition to the TGF-β-induced tumor suppressor pathway.

Nat. Neurosci. 5:2002;946-954 The authors describe how Runx3 controls the target specific pathfinding of proprioceptive DRG neurons in the spinal cord. Although proprioceptive ganglion neurons do not reach central and peripheral targets, they do not seem to undergo apoptosis unexpectedly. As TGF-β is considered to regulate ontogenetic neuron death, Runx3 might be involved in this process. Because neuroblastoma cells and DRG neurons originate from neural crest, the results obtained by this study might contribute to the study of neuroblastoma, especially in the light of [26].

U.S.A. 99:2002;13073-13078 The transcriptional coactivator p300 is targeted by viral oncoprotein and is suggested to be a tumor suppressor. The transcriptional response to TGF-β was severely impaired in the cancer cells that contain mutations in the p300 gene accompanied by the inactivation of the second allele. They show that Smad signaling and p300, but not the mutant forms identified in cancer cells, synergistically activate the p21 gene promoter, suggesting that the growth-suppressive effect of TGF-β may be mediated by this mechanism.

The authors find that this is caused by incomplete angiogenesis itself, caused by a lack of hematopoietic stem cells that secrete angiopoietin-1, which is required for angiogenesis.
