    • Like JAMM, metal-dependent peptidases often possess two conserved acidic residues: one that acts as a ligand for zinc and a second that acts as a nucleophile to promote attack by water on the peptide backbone. Based on the common occurrence of Glu as a nucleophile and on the predicted secondary structure of the Jab/MPN domain, we propose that the Glu helps polarize a water molecule for hydrolytic attack and the COOH-terminal His and Asp residues serve as three ligands for zinc.
