(2002) [12••] provide very suggestive evidence that a second exocytic pathway emanates from prevacuolar endosomes. This is the best evidence so far that transport is possible directly from endosomes to the plasma membrane in yeast and is all the more surprising because prevacuolar endosomes have been considered late endosomal compartments.

It also shows that the ubiquitinated protein binds to a complex containing Vps23p, and that a vps23 mutation that blocks this binding prevents entry. This is a first step in the biochemical analysis of MVB formation.

The authors clearly demonstrate that HIV budding requires at least some of the same machinery as the topologically equivalent formation of multivesicular bodies, even though it occurs on a different membrane. This is potentially a good system to study this mysterious process. Also, the authors provide an early demonstration of the power of RNA interference technology in studying protein function in mammalian cells.

Dev Cell. 2:2002;283-294. The authors provide elegant genetic evidence for a role of the yeast AP1 adaptor in the sorting of a particular protein. They provide suggestive evidence that AP1 is required for retrieval from endosomes, although it is possible that the effect is instead - or also - on export from the Golgi. It is notable because this is the most specific sorting defect found so far for AP1 in yeast.

This paper shows that Hrs, clathrin and ubiquitinated proteins co-localise in patches on early endosomal membranes, and that overexpression of Hrs directs an artificially ubiquitinated form of the transferrin receptor into these patches, inhibiting its recycling to the cell surface. Together with Sachse et al. (2002) [78•], this paper provides strong evidence that clathrin is involved in sorting ubiquitinated proteins from early to late endosomes.
