    • Physiological analysis of transgenic Arabidopsis thaliana plants expressing one nitrilase isoform in sense or antisense direction
    • This paper examines the role of nitrilase in indole-3-acetic acid biosynthesis in Arabidopsis. It suggests that synthesis of indole-3-acetonitrile (IAN), the substrate for nitrilase, may be limiting. Expression of one of the four nitrilase genes (NIT2) was strongly induced by its substrate, IAN.

    • IAR3 encodes an auxin conjugate hydrolase from Arabidopsis
    • Indole-3-acetic acid (IAA)-amino-acid conjugates are hydrolysed to free IAA in plant tissues by hydrolases that are specific for particular amino acid conjugates. The IAR3 (IAA-Ala-resistance) gene was isolated and shown to encode a hydrolase with specificity for IAA-Ala and IAA-Gly. CaMV35S-mediated expression in Arabidopsis of IAR3 and ILR1, which is specific for IAA-Leu and IAA-Phe, resulted in plants with increased sensitivity to IAA-Ala and IAA-Leu/Phe, respectively, in terms of inhibition of root growth

    • This paper provides a good example of the use of genetic manipulation to study hormone function. The content of zeatin-type cytokinins was increased in Arabidopsis by heat-induced expression of the ipt gene. The transgenic plants contained higher levels of transcripts for the KNAT1 and KN1 homeobox genes. On the basis of this and other evidence, the authors suggest that cytokinins act upstream of these genes in maintenance of the shoot meristem

    • The Arabidopsis homeobox gene KNAT1 was expressed in lettuce under control of the pea plastocyanin promoter. The resulting morphological changes were consistent with elevated cytokinin concentration, which was confirmed by immunocytological analysis. The result suggests a possible role for homeobox genes in regulating cytokinin biosynthesis
