Antiserum to xPin1 or serum from nonimmunized rabbits was incubated with Affi-prep protein A (Bio-Rad Laboratories, Hercules, CA), phosphate-buffered saline, and bovine serum albumin (2 mg/ml) (Sigma) at 1:4:25 (protein A was measured by volume of slurry as purchased). The supernatant was removed, and the extract was exposed to a fresh tube of the bound after 40 min of rotating at 4°C.

38. Recombinant baculoviruses encoding His-tagged human cyclin B1s were gifts from D. O. Morgan. Purification was carried out as described by Kumagai and Dunphy (34). Each extract required titration with cyclin B to determine the appropriate concentration for observing effects on cell cycle timing.
