    • Synaptic activation causes the mRNA for the IEG Arc to localize selectively near activated postsynaptic sites on dendrites
    • The authors demonstrate that the mRNA for the immediate-early gene Arc is targeted to activated synapses in the hippocampus following tetanic stimulation of the afferent fibers in the perforant path. The authors use the laminar pattern of innervation from the perforant path onto dendrites in the hippocampus to stimulate only a subset of synapses. By in situ hybridization they demonstrate that Arc mRNA is selectively localized to the dendritic laminae that received the tetanic input. The ability to selectively target mRNAs (as well as proteins) to activated synaptic regions is likely to be crucial for the specificity of synaptic plasticity.

    • The authors demonstrate that synaptic activation leads to an increase in CaMKII protein in dendrites; this increase is dependent upon local (dendritic) protein synthesis. In a hippocampal slice preparation, an increase in CaMKII protein is induced by tetanic stimulation of the Schaffer collateral pathway and is blocked by translation inhibitors. The increase can be detected as early as 5 min post-stimulation.

This is the first proposed molecular mechanism describing how mRNA translation may be regulated in the dendrite.
