This cell line was then transfected with HeLa cell polythymine-primed cDNA library cloned in-frame with the GAL4 activation domain (Clontech, Palo Alto, CA), and 3 million transformants (about three times the library complexity) were assayed for a positive two-hybrid interaction on histidine-deficient plates, followed by confirmation with a β-galactosidase filter assay. Positive clones encoded fusion proteins of the Gal4 activation domain with MKK1 (J98, initiating at glycine 61), MKK2 (J31, J62, and J22, initiating at glycine 29, glycine 29, and glutamine 32, respectively), and MKK4 (J27, initiating at glutamine 38).
