The selection methodology employed was the 'selectively infective phage' (SIP) technology, a method in which ligand binding of the displayed polypeptide is directly linked to phage infectivity. The selected scFv fragment showed an increased thermodynamic stability compared with the competing sequences.

This is achieved with a phagemid/helper phage system by engineering a protease site into wild-type g3p encoded by the helper phage. Upon protease treatment, N1 and N2 are cleaved off. Therefore, the only N1 and N2 domains, which are needed for infection, can come from the fusion proteins, provided that it is itself resistant to proteolysis.

A novel phage display selection for enzymatic activity is described using staphylococcal nuclease as a model system. The enzyme and the substrate were displayed on the same phage particle. This was achieved by fusing the enzyme directly to g3p and fusing a coiled-coil peptide to another copy of g3p, which heterodimerizes with another coiled-coil peptide that is chemically linked to the substrate, a biotinylated oligonucleotide. After the binding of the phages to streptavidin beads through the biotin moiety, phages displaying active enzymes were released by cleavage of the oligonucleotide in a reaction on the same phage particle. This methodology has the potential to be used for the selection and evolution of those protein catalysts that can be displayed.

Enzyme-calmodulin fusion proteins were displayed on filamentous phage. A substrate linked to a calmodulin-binding peptide could then be anchored in proximity to the enzyme. Phages displaying active enzymes were selected by product binding. This may be a general methodology for the isolation of enzymatic activities from libraries of proteins that can be displayed.
