CREB-mediated expression of a CRE-linked reporter transgene was increased in the CA1 region of the hippocampus following contextual fear conditioning. CREB-mediated transcriptional activation has been implicated in long-term memory formation in many systems and this is a clear demonstration of learning-induced activation of CREB-linked transcription.

    • Moser El, Krobert KA, Moser MB, Morris RG: Impaired spatial learning after saturation of long-term potentiation. Science 1998, 281:2038-2042. If LTP is important for memory formation then producing LTP at all synapses in one part of a circuit where memory is stored should impair the storage. This paper demonstrates such an impairment in water maze performance with saturation of LTP at perforant path synapses to the hippocampus.

    • Xu L, Anwyl R, Rowan MJ: Spatial exploration induces a persistent reversal of long-term potentiation in rat hippocampus. Nature 1998, 394:891-894. LTP that has been electrically induced in the CA1 region of the hippocampus is reversed when the rats are placed in a novel environment. This surprising finding suggests that a mechanism exists to reset recently potentiated synapses when an animal needs to learn new information.

J Neurosci 1998, 18:4177-4188. Transgenic mice are generated in which an embyronic subunit of the NMDA receptor is expressed in adult mice. This resulted in altered NMDA currents and reduced LTP to 30% of wild-type levels. No impairment in complex spatial learning was found, suggesting that LTP can be substantially impaired without impacting on learning.

Nature 1998, 394:577-581. Mice with a targeted deletion in a gene encoding a receptor for an endogenous peptide associated with pain perception are found to learn faster than wild types on two different tasks. There is also a slight enhancement in LTP. This is the first report of a genetically modified mouse with enhanced learning ability although the mechanism by which the enhancement is achieved is not clear.

Nature 1998, 396:433-439. A mouse with a deletion in the PSD-95 gene is shown to have enhanced LTP and to have lost the capacity for long-term depression (LTD). The animals are severely impaired in spatial learning. The findings demonstrate an interesting regulatory function for PSD-95 in controlling the balance between LTP and LTD, presumably by regulating the clustering of signaling molecules in close proximity to NMDA receptors. In addition, the results demonstrate that enhancing LTP will not necessarily improve memory ability.
