Chimeric molecules were created by broad range amplification of DNA extracts containing multiple bacterial species. The authors believe that the presence of such molecules can compromise the integrity of sequence data-bases and cause misinterpretation of the phylogenetic status by the creation of false genetic diversity within prokaryotes.

Sodium polyanetholsulfonate (SPS) is a water soluble, high molecular weight molecule that is inhibitory to PCR. It is commonly added to blood cultures as an anticoagulant and to inactivate complement. SPS generally co-purifies with DNA and severely limits the sensitivity of PCR amplification from blood cultures. The authors found that extraction of DNA with benzyl alcohol removed SPS and improved PCR sensitivity.

Bacterial strains that were identified as members of the Mycobacterium avium complex were further characterized by automated DNA sequence analysis of a 360 bp portion of the hsp65 gene. The authors found that the level of variability exhibited by this molecular target allowed the unambiguous discrimination of 21 clinical isolates and 35 reference strains.

J Infect Dis 1998, 178:776-782. The authors further demonstrated the usefulness of the assay described by Swanson et al. [29•] by characterizing Mycobacterium avium complex isolates from 136 children. In addition to speciation of these isolates, they found that the predominant species associated with mycobacterial infection in HIV infected children was M. avium, whereas M. intracellulare was most often responsible for mycobacterial infections in children not infected with HIV.

The isolates were categorized into two major divisions and nine subdivisions. Significant nucleotide polymorphisms were found to be unique to specific geographic regions.

A new Corynebacterium species was isolated from a human sputum specimen, characterized and described. The 16S rDNA sequence of this isolate was compared with other prokaryotes and was found to be most closely related to, but quite divergent from, other members of the genus Corynebacterium.

The authors describe a novel bacterial species most closely related to the genus Corynebacterium. Although phenotypic methods characterized this isolate as a Corynebacterium, no species-level discrimination could be made. Following 16S rDNA analysis, the isolate was determined to be a new species of this genus, with >92% sequence similarity.

The isolate was found to be most closely related to Corynebacterium genitalium.

Gastroenterology 1998, 114:755-763. This was an epidemologic study that was conducted to determine if Helicobacter infection is associated with cholecystitis. By using Helicobacter specific PCR primers, the authors amplified DNA from bile fluid and gallbladder tissues. Subsequent DNA sequencing indicated multiple Helicobacter sp. among the various clinical samples.

J Med Microbiol 1998, 47:695-704. The authors used 16S rDNA PCR and sequencing to detect Helicobacter DNA in multiple gastric biopsy samples, including several normal controls. The conclusion was that even though this method was sensitive enough to amplify such targets from gastric biopsies, no correlation could be made between the presence of Helicobacter DNA and clinical infection.
