Sodium polyanetholsulfonate (SPS) is a water soluble, high molecular weight molecule that is inhibitory to PCR. It is commonly added to blood cultures as an anticoagulant and to inactivate complement. SPS generally co-purifies with DNA and severely limits the sensitivity of PCR amplification from blood cultures. The authors found that extraction of DNA with benzyl alcohol removed SPS and improved PCR sensitivity.

[29•] by characterizing Mycobacterium avium complex isolates from 136 children. In addition to speciation of these isolates, they found that the predominant species associated with mycobacterial infection in HIV infected children was M. avium, whereas M. intracellulare was most often responsible for mycobacterial infections in children not infected with HIV.

Gastroenterology 1998, 114:755-763. This was an epidemologic study that was conducted to determine if Helicobacter infection is associated with cholecystitis. By using Helicobacter specific PCR primers, the authors amplified DNA from bile fluid and gallbladder tissues. Subsequent DNA sequencing indicated multiple Helicobacter sp. among the various clinical samples.

J Med Microbiol 1998, 47:695-704. The authors used 16S rDNA PCR and sequencing to detect Helicobacter DNA in multiple gastric biopsy samples, including several normal controls. The conclusion was that even though this method was sensitive enough to amplify such targets from gastric biopsies, no correlation could be made between the presence of Helicobacter DNA and clinical infection.
