This paper reveals the conformation of a HEAT-motif-containing protein, the position of conserved hydrophobia residues that are likely to form the protein - protein interaction surface and the structural consequences of point mutations in the forms of the PR65/A subunit that are associated with lung and colon tumours. Conformational flexibility within the molecule is described.

Protein - peptide interactions utilise conserved asparagine and tryptophan residues on adjacent armadillo repeats. Compare this paper to [20••], which describes differences between the conformation of the NLS bound to the ankyrin repeats of IκBα and the armadillo motifs of importin α.
