This study evaluates the survival requirements of a clone of T cells of which the class I MHC restriction is known. This clone, recovered before or after stimulation with antigen in vivo, is transferred to mice deficient in various MHC class I molecules. The results obtained allow the direct correlation of cell survival with particular TCR-MHC interactions, as well as the direct comparison of survival requirements of the naive and memory T cells expressing the same TCR specificity.

J Exp Med 1998, 188:71 -82. References [37••-39••,40•,41•,42••] follow antigen-specific T cells in vivo, using their ability to bind specific antigenic peptides that are coupled to MHC tetramers. This methodology, developed in the laboratory of Mark Davis, represents a major breakthrough that allows us, for the first time, to follow clonal evolution during immune responses. All studies show that the relative representation of different antigen-specific T cell clones found in the peak of the immune response is the same as that found after the establishment of the memory T cell pool. These data do not support the hypothesis that survival of memory T cells is dependent on the recognition of cross-reactive peptides because, if this was the case, selection of particular clones would be expected.
