This paper described axonal loss (up to 82%) in multiple sclerosis lesions from 11 individuals with clinically silent multiple sclerosis. These individuals had no reported neurologic symptoms during their lifetimes. The lack of neurologic symptoms were attributed to the following: most lesions were in clinically silent areas; low levels of total axonal loss; neuronal redundancy of affected areas; and remyelination.

This paper described axonal degeneration in mice that lack the myelin-associated glycoprotein. These mice myelinate normally and develop late-onset axonal pathology and degeneration. These observations support the hypothesis that myelin provides external trophic signals to axons. Loss of this trophic effect in multiple sclerosis may lead to degeneration of chronically demyelinated axons.

This paper describes axonal degeneration in mice that lack proteolipid protein. These mice form normal amounts of myelin membrane and begin to develop axonal swellings at 6-8 weeks of age. By 12 months of age, axonal degeneration in the absence of primary demyelination occurs in fiber tracts with axonal swellings. These data support the possibility that chronic demyelination in multiple sclerosis can lead to axonal degeneration.
