30,31 Alkoxyamino esters were used to synthesize vancomycin conjugates 5d-e and 6d-e containing alkoxy linkers. Aglycone derivatives were prepared by reacting the aglycone (1 equiv) with 5 equiv dicyclohexylcarbodiimide, 5 equiv of the amine, 2 equiv of hydroxybenztriazole, and 2 equiv of triethylamine or diisopropylethylamine in anhydrous DMSO (20 mL/mmol).
