    • 2+ (6). DNA (5 ng) was digested with Hae III, followed by 10 cycles of PCR amplification with primers DG88 and DAR5 (15). One-tenth of the amplified product was digested with Hae III (to remove any Hae III-sensitive material that survived the initial digestion), followed by 20 cycles of PCR with the same primers. Subsequently, PCR products were digested with Hha I, separated on a 6% polyacrylamide gel in tris-borate-EDTA (TBE) buffer and blotted onto Hybond N+. Blots were hybridized with oligonucleotide NEB1233, and products were visualized by autoradiography.
