Farnesyl diphosphate synthase catalyzes a carbocation-mediated isoprenoid chain elongation reaction, forming farnesyl diphosphate, which, in turn, is the substrate of a sesquiterpene cyclase. Farnesyl diphosphate synthase provided the first example of the core terpenoid synthase α-barrel fold subsequently observed in the structures of pentalenene synthase, 5-epi-aristolochene synthase and aristolochene synthase.

Nevertheless, a comparison of these 'un-natural' cyclases with naturally occurring terpenoid cyclases highlights the relative ease with which biology accommodates highly reactive electrophiles.
