A mutation in the silencing gene SIR4 had previously been shown to extend life span [10], and was hypothesized to cause the relocalization of the Sir complex to a locus involved in life span regulation. This paper identifies this locus as the ribosomal DNA. It also describes two genes required for movement of the Sir complex to the nucleolus.

SGS1 is the yeast homolog of the WRN gene, a mutation in which causes premature aging in man. It is shown that sgs1 mutant cells have a decreased longevity and display phenotypes of old cells early in their life span. Nucleolar fragmentation is described as a new marker of old age in yeast cells.

It shows that circular rDNA species accumulate in mother cells as they divide and eventually cause their death. It is shown that Sir4p, like Hdf 1 p, is required for DNA repair by nonhomologous end-joining, together with Sir2p and Sir3p. Consistently, mutations in SIR2, SIR3 or SIR4 increase the radiation sensitivity of a rad52 mutant strain.
