Unique requirements for retinoid dependent transcriptional activation by the orphan receptor LXR

of outstanding interest. Similar to the findings reported in [22], this paper characterizes the unique mechanism whereby the binding of ligand to one receptor (RXR) induces a conformational change in its unliganded partner (LXR) that results in transactivation. The ability of the RXR/LXR heterodimer to be transactivated by one or both receptors' ligands is a unique feature of this subclass of heterodimer.

Cholesterol and bile acid metabolism are impaired in mice lacking the nuclear oxysterol receptor LXRα
-
of outstanding interest. The knockout of the mouse LXRα gene demonstrates definitively that LXRα functions as a key sensor of dietary cholesterol and regulates the elimination of cholesterol from the body. In perfect symmetry to the SREBP pathway that controls the feedback mechanism that represses the rate-limiting step in cholesterol synthesis, LXRα is shown to regulate the feed-forward pathway that induces the rate-limiting step in the catabolism of cholesterol to bile acids.
