This article describes the structural characteristics of human bactericidal/permeability increasing protein (BPI) at a crystal structure resolution of 2.4A. BPI is closely related to lipopolysaccharide-binding protein (LBP) and alignment of the amino acid sequences shows that structurally important residues are conserved. BPI was shown to interact with the acyl chains of phosphatidylcholine and it was postulated that both BPI and LBP interact with the acyl chains of lipopolysaccharide. This provides the first evidence of those structural components of lipid A that are recognized by LBP.

Infect Immun 1997, 65:293-297. This article was the first to identify single point mutations in CD14 that have significant effects on its ability to bind to lipopolysaccharide (LPS). Specific charge reversal mutations had the greatest effect on LPS binding, pointing out the importance of charge in CD14 recognition of LPS. In addition, it was shown that CD14 can selectively recognize two different LPS species. Although this has therapeutic potential, additional data (RP Darveau, J Bajorath, C Seachord, MC Cunningham, unpublished data) suggest that CD14 presents a charged surface for LPS binding and the importance of specific charge interactions remains unknown.

J Biol Chem 1997, 272:19688-19696. These authors constructed the first genetically engineered strain with an altered lipid A fatty acid composition. The authors were able to complement an lpxA mutation in E. coli with the lpxA gene from Neisseria meningitidis. The resultant E. coli strain was viable yet the lipid A contained the shorter fatty acids characteristic of N. meningitidis. This approach to the study of lipid A function in bacteria will be invaluable in the study of lipid A function in host interactions.

Mutants with decreased ability to stimulate E-selectin expression from endothelial cells were identified. A mutation in the msbB gene resulted in the production of lipopolysaccharide that only lacked myristate, identifying the importance of this secondary fatty acid for host cell recognition. This was the first report of a lipid A mutation that was not conditionally lethal for growth and was used to confirm the significant role of lipid A when viable bacteria are presented to host defense cells.

J Biol Chem 1996, 271:25898-25905. This article describes an increase in the specific activity of LpxC when the activity of LpxA is inhibited. An increase in the specific activity of LpxC, the first committed step in lipid A synthesis, indicates that E coli may be able to regulate the amount of lipid A synthesized in response to the lipid A content of its outer membrane. Several different innate host defense components act by disrupting the lipopolysaccharide barrier of the outer membrane and this article provides a means for testing the hypothesis that E. coli may respond to host attack by increasing lipid A synthesis.

This article identified Yersinia enterocolitica bacterial chromosomal genes that were preferentially expressed after intragastric ingestion in mice. One gene was found to encode a protein with homology to LpxA from Y. enterocolitica, yet was distinct from other characterized acyltransferases. These data remain speculative but suggest that Y. enterocolitica may contain an lpxA analogue which is activated in the in vivo environment and alters the lipid A fatty acid composition.

This article examined the lipid A composition of Salmonella typhimurium strains with mutations in the virulence genes phoP-phoQ. It was found that mutations in the phoP-phoQ system, which regulates numerous genes in response to host microenvironments, induced structural modifications of lipid A. These modifications altered lipopolysaccharide-mediated expression of E-selectin by endothelial cells and tumor necrosis factor-α expression by adherent monocytes. These data provide the strongest evidence to date that lipid A may be altered in response to host microenvironments.
