This paper presents the first evidence that α-MSH inhibits renal injury from ischemia/reperfusion, even when first started 6 h into the reperfusion period. α-MSH inhibited maladaptive inflammatory and NO cascades initiated during the reperfusion period.

The authors proposed that α-MSH could be a new treatment of traumatic spinal cord injury.

This is an immunohistochemistry study using rabbit polyclonal antibodies to melanocortin-1 and melanocortin-5. Melanocortin-1 receptors were expressed on neutrophils and apical membranes of collecting ducts. Melanocortin-1 was also detected by protein blotting on mouse cortical tubule cells. Neutrophils also express melanocortin-5.

Kidneys were perfused ex vivo without neutrophils, subjected to ex vivo ischemia, then reperfused ex vivo without neutrophils. α-MSH protected glomerular and tubular function, suggesting that it has direct effects on the kidney independent of neutrophil-mediated tissue injury.
