Molecular mechanism of desensitization of the chemokine receptor CCR5: Receptor signaling and internalization are dissociable from its role as an HIV-1 co-receptor
-
Aramori I, Zhang J, Fergusson SS, Bieniasz PD, Cullen B, Caron MG. Molecular mechanism of desensitization of the chemokine receptor CCR5: receptor signaling and internalization are dissociable from its role as an HIV-1 co-receptor. EMBO J 1997; 16:4606-4616. This paper describes experiments that show that, in response to chemokines, CCR-5 is desensitized, phosphorylated, and sequestered only after overexpression of G-protein coupled receptor kinases and β-arrestins. However, the function of CCR5 as an HIV coreceptor is unaffected.

Selective expression of the eotaxin receptor CCR3 by human T helper 2 cells
-
Sallusto F, MacKay CR, Lanzavecchia A. Selective expression of the eotaxin receptor CCR3 by human T helper 2 cells. Science 1997; 277:2005-2007. This paper shows for the first time that a chemokine receptor is selectively expressed on T-cell subsets. An antibody to CCR3 was used to isolate T cells from peripheral blood that give rise to Th2-polarized cell lines and to identify Th2 cells derived from naive T cells in vitro.

RANTES and monocyte chemoattractant protein-1 (MCP-1) play an important role in the inflammatory phase of crescentic nephritis but only MCP-1 is involved in crescent formation and interstitial fibrosis
-
Lloyd CM, Minto AW, Dorff ME, Proudfoot A, Wells TNC, Salant DJ, Gutierrez-Ramos J-C. RANTES and monocyte chemoattractant protein-1 (MCP-1) play an important role in the inflammatory phase of crescentic nephritis but only MCP-1 is involved in crescent formation and interstitial fibrosis. J Exp Med 1997; 185:1371-1380. This study involved blocking the functions of MCP-1 or RANTES during development of crescentic nephritis using neutralizing antibodies or a RANTES receptor antagonist. Evidence is provided to show that MCP-1 is critical, not only in the accumulation of leukocytes in the kidney, but also in the progression of disease.
