CTLA4-lg and anti-CD40 ligand prevent renal allograft rejection in primates
-
of special interest. This study showed that costimulation blockade prolongs renal allograft survival in rhesus monkeys. Two animals treated in such a manner experienced late rejection, but a repeat course of the induction regimen successfully restored normal graft function. Unexpectedly, lymphocytes from rejection-free animals responded vigorously in mixed lymphocyte reactions to donor, and to third-party, cells.

Preclinical studies of allograft tolerance in rhesus monkeys: A novel anti-CD3-immunotoxin given peritransplant with donor bone marrow induces operational tolerance to kidney allografts

Chimerism was observed in all immunotoxin-treated animals, including those that did not receive DBM; however, the level of chimerism did not correlate with graft survival.

of special interest. This study demonstrated that CD154 is expressed not only on T cells but also on activated platelets. This finding indicates that the effects of CD154 blockers may not be limited to downregulating T cell function, but that the function of platelets may have also been modulated by these blockers. This suggests that platelets may play an important role in allograft rejection.

In utero hematopoietic stem cell transplants prolong survival of fetal kidney transplantation in monkeys
-
of special interest. This study examined the potential for donor bone marrow (BM) transplantation to induce tolerance in utero. T-cell-depleted paternal BM was injected in utero into the peritoneal cavity of two female fetal rhesus monkeys at day 61 of gestation. Chimerism was detected as early as six weeks after birth and until two years of age. In contrast to controls, chimeric monkeys had prolonged graft survival and no clinical evidence of graft failure for at least seven weeks; however, biopsies performed one - four weeks post-transplant showed signs of moderate/acute rejection. Consistent with these in vivo findings, in vitro mixed lymphocyte reactions to the donor cells were reduced compared with responses to third-party cells.
