These data could potentially explain how alterations in tuberin lead to the tuberous sclerosis complex phenotype.

The Eker rat was developed as a model for hereditary renal cell carcinoma. It was found that these rats carry a germline mutation of the TSC2 gene. Further studies revealed that these rats develop nervous system lesions similar to the giant cell astrocytomas and hamartomas seen in tuberous sclerosis complex.

Clin Cancer Res 1997, 3:473-478. The use of genetic markers to predict prognosis and treatment outcomes is becoming an important part of standard medical practice. This paper, by presenting results in conflict with earlier studies, points out the need for large and extensive studies prior to using a genetic test in the clinical situation.

    • Raffel C, Jenkins R, Fredenck L, Hebrink D, Alderete B, Fults D, James C: Sporadic medulloblastomas contain PTCH mutations. Cancer Res 1997, 57:842-845. The patched gene has been extensively studied in Drosophila, where it functions as a developmental regulator. The human homolog PTCH functions in a similar manner. The finding that PTCH is mutated in some medulloblastomas suggests potential etiologic mechanism of tumorigenesis.
