    • Prostaglandin F2-like compounds, F2-isoprostanes, are present in increased amounts in human atherosclerotic lesions
    • 2 alpha compared with human umbilical vein, which had 11 pg/mg dry weight. Hydroxy linoleic acid was present at much higher levels, i.e. 0.5% of the total linoleic acid. No hydroxy linoleic acid was detected in human umbilical vein. It was concluded that, for the evaluation of lipid peroxidation in lesions, hydroxy linoleic acid is more suitable than isoprostane.

    • 2 alpha generation during coronary reperfusion. A potential quantitative marker of oxidant stress in vivo
    • 2 alpha. Circumflex artery occlusion in a canine model did not increase urine levels. However, levels were significantly increased after reperfusion. At 30 min after lytic therapy, levels were threefold greater than pre-intervention levels but returned to normal at 24 h. Similar changes were seen in patients given thrombolytic therapy.

    • Increased formation of the isoprostanes IPF2alpha-I and 8-epi-prostaglandin F2 alpha in acute coronary angioplasty: Evidence for oxidant stress during coronary reperfusion in humans
    • 2 alpha were increased twofold 6 h after angioplasty. After 24 h, levels had returned to those observed pre-intervention. Thrombolytic therapy also increased isoprostane production; however, there was only a slight effect of angiography.

    • The human thromboxane A2 receptor alpha isoform (TP alpha) functionally couples to the G proteins Gq and G11 in vivo and is activated by the isoprostane 8-epi prostaglandin F2 alpha
    • 2 alpha stimulated calcium flux in cells co-transfected with either Gq or G11 but not with thromboxane receptor alone. Treatment of platelets with thromboxane antagonist desensitized iso F2 stimulation. The authors demonstrate that 8 iso PGF2 alpha acts on the thromboxane receptor by a Gq or G11 coupled mechanism.

The authors demonstrate that incubation of LDL with fibroblasts overexpressing 15-lipoxygenase generate MM-LDL. The MM-LDL are shown to have direct chemotactic activity for monocytes. Bioactivity resided in the lipid fraction, and was blocked by use of PAF-receptor antagonists. These studies suggest yet another proinflammatory activity of oxidized phospholipids is generated when LDL is mildly oxidized.

In this study, in lupus patients who have increased titers of antiphospholipid antibodies, it was shown that there was a very strong correlation between the antibody titer and the urinary excretion of isoprostanes. This study supports the hypothesis that antiphospholipid antibodies may form in response to pro-oxidative conditions.
