This gene-mapping study identifies chromosomal segments where synteny is conserved between zebrafish and human. Analysis of the conserved segments suggests that two genomic duplications occurred in vertebrate evolution prior to the divergence of zebrafish and human ancestors, and that a third whole-genome duplication occurred in the zebrafish lineage after divergence. The observation that some linkage relationships have been preserved since the divergence of zebrafish and human raises the possibility that the locations of zebrafish genes can be predicted by comparison with human maps.

Ectopic expression studies show that FGF-8 can expand lateral fates at the expense of ventral, supporting a role in dorsal-ventral patterning.

    • Amacher S, Kimmel C: Promoting notochord fate and repressing muscle development in zebrafish axial mesoderm. Development 1998, 125:1397-1406. Analysis of flh;spt double mutants provides genetic evdience that the notochord-specific flh gene functions to repress spt, which is required for development of trunk muscle. Transplantation studies show that cells mutant for flh, which encodes a homeodomain protein, can acquire a notochord fate in response to signals present in a wild-type environment.

    • Zhang J, Talbot WS, Schier AF: Positional cloning identifies zebrafish one-eyedpinheadas a permissive EGF-related ligand required during gastrulation. Cell 1998, 92:241-251. These authors employ a positional cloning approach to identifiy a new gene with essential function in prechordal plate and endoderm development. The oep gene product might be a previously unrecognized component of a known signalling pathway, or it might define a novel pathway operating in the early embryo.

    • •] have constructed a Tc1-class transposon by correcting inactivating mutations from salmonid transposon- like sequences and showed that this synthetic element, Sleeping Beauty, is active in fish and mammalian cells. These reports lay the foundation for transposon-mediated gene transfer and, if transposition efficiency can be improved, mutagenesis in zebrafish.

The authors use laser-activated caged-fluorescein fate mapping to define the locations of cardiac progenitors in the embryo and to identfiy the source of cells that can divert to a cardiac fate when the normal progenitors are ablated. This paper represents an important advance in the utility of caged compunds in the transparent zebrafish embryo.
