This report analyses TCR α, β, γ and δ gene rearrangements, as well as the expression of the recombination enzymes (recombination activation genes [RAGs] 1 and 2, Ku80 and terminal deoxynucleotidyl transferase), in the line of IL-7Rα KO mice with the milder defect in T cell development. The results indicate a specific absence of TCR γ V-J recombination, with all other TCR gene rearrangements and the expression of recombination enzymes being detectable at normal levels in thymocytes from this line.

of special interest. This report describes the absence of NK cells and a reduction in intestinal intraepithelieal lymphocytes (IELs) in IL-2Rβ KO mice. Both TCRαβ CD8αα and TCRγδ IELs are reduced in these mice. These data indicate a role for IL-2 or IL-15 in the development of multiple subsets of extrathymic lymphocytes.
