    • v mice were bred with mice containing a transgene BCR specific for lysozyme, and B cells were isolated from the progeny. These B cells were hyper-responsive to antigen triggering, suggesting that wild-type HCP is a terminator of B-cell signaling. Mixed bone marrow experiments, using HCP-deficient cells and lysozyme antigen with lower valency, provide evidence that tolerance thresholds are set by the strength of BCR signaling.

Demonstrates that FcγRIIB1-mediated inhibitory signaling involves recruitment of HCP to activated (i.e. tyrosine phosphorylated) FcγRIIB1, and that binding of HCP to FcγRIIB1 is likely to stimulate HCP PTPase activity. Binding and activation are dependent on a 13 amino acid region within FcγRIIB1. Analysis of FcγRIIB1 signaling in B cells isolated from HCP-deficient me mice confirms that HCP facilitates FcγRIIB1 inhibition of BCR signaling.

Demonstrates that HCP is recruited to activated CD22 following BCR ligation, that binding to CD22 is likely to stimulate the PTPase activity of HCP, and that ligation of CD22 to prevent co-aggregation with the BCR significantly lowers the threshold of BCR activation. The authors suggest that interactions of CD22 with its counter-receptors on T cells may remove HCP from the BCR complex, and thus allow BCR signaling to occur in microenvironments that contain less antigen.

These findings convincingly argue that HCP functions to terminate erythropoietin proliferative signaling by dephosphorylating JAK2.

Demonstrates that LAR and the LAR interacting, coiled-coil protein LIP.1 colocalize at the ends of focal adhesions most proximal to the nucleus and are excluded from the distal ends of focal adhesions, thus localizing to sub-regions of focal adhesions that are presumably undergoing disassembly. (Focal adhesions have been observed to translocate without loss of attached stress fibers; thus, polarized assembly [at the distal end] and disassembly [at the proximal end] of focal adhesions probably occur.) This localization suggests that LAR may function in part to regulate the length of focal adhesions. LIP.1 binds to the LAR membrane-distal D2 PTPase domain and may function to recruit LAR to focal adhesions. The LAR-LIP.1 complex may represent an independent matrix-cytoskeletal linkage with structural and signaling roles.
