The inclusion of at-risk individuals permits discrete survival curves to be drawn for each CAG sizes. The implications for predictive testing are discussed.

Cell 1996, 87:493-506. The authors describe an intriguing neurological phenotype in mice transgenic for the first exon of Hdh with very large CAG lengths. The phenotype of these mice includes motor abnormalities such as tremor and involuntary movements, drastic weight loss and reproductive anomalies, but does not include specific neurodegeneration.

The authors show that despite differences to the murine protein, human huntingtin is expressed in a identical manner to that of the murine protein and can rescue the embryonic lethal phenotype in mice homozygous for a targeted disruption in murine Hdh exon 5.

Because there is a subtle increase in the rate of cleavage for huntingtin with long CAG tracts, this study suggests a possible link between CAG expansion and initiation of apoptosis.

This landmark study illustrates the crucial role for caspase 3 (apopain) in developmental neural apoptosis. The authors show that mice deficient in apopain have grossly abnormal neural ectopic masses that are large enough to cause secondary skull defects. Importantly, the overall neural organization of these ectopic masses appears normal.
