Regulation of sucrase-isomaltase gene expression in human intestinal epithelial cells by inflammatory cytokines
-
Ziambaras T, Rubin DC, Perlmutter DH: Regulation of sucrase-isomaltase gene expression in human intestinal epithelial cells by inflammatory cytokines. J Biol Chem 1996, 271:1237-1242. Two typical features of small intestinal inflammation are crypt hyperplasia and villus atrophy. One opinion is that the epithelium becomes populated with relatively undifferentiated enterocytes, leading to a net reduction in brush border hydrolases. On the other hand, this paper reports that cytokines directly affect sucrase-isomaltase expression. They survey multiple cytokines and conclude that interleukin-6 and interferon gamma specifically downregulate sucrase-isomaltase; understanding the in vivo relevance of autocrine interleukin-6 in relation to this finding will be interesting.

-
Reinecker HC, MacDermott RP, Mirau S, Dignass A, Podolsky DK: Intestinal epithelial cells both express and respond to interleukin 15. Gastroenterology 1996, 111:1706-1713. A comprehensive analysis of human and rat IEC lines, as well as freshly isolated cells, showing interleukin-15 mRNA protein. They also show that phosphorylation takes place after binding of interleukin-15 to the cells, signifying that the receptors are functional. Also, this group cloned (by polymerase chain reaction) and report the sequence of rat interleukin-15 in the paper.

Intestinal epithelial cell line induction of T cell differentiation from bone marrow precursors
-
Marie D, Kaiserlian D, Croitoru K: Intestinal epithelial cell line induction of T cell differentiation from bone marrow precursors. Cell Immunol 1996, 172:172-179. This paper provides direct evidence for IEC support of T-cell maturation by coculturing a mouse IEC line with lymphocyte-depleted mouse bone marrow and showing that cells bearing T-cell markers grow out of the mix. They also show that the effect is mediated by soluble factors, which remain to be identified.

Pattern of proinflammatory cytokine mRNA expression during Trichinella spiralis infection of the rat
-
Stadnyk AW, Kearsey JA: Pattern of proinflammatory cytokine mRNA expression during Trichinella spiralis infection of the rat. Infect Immun 1996, 64:5138-5143. This may be the first description of interleukin-1β made by small intestinal epithelial cells. The issue of whether contaminating IEL in the cell preparations produced significant levels of interleukin-1β was ruled out by enriching for lymphocytes and showing that the interleukin-1β mRNA signal copurified with the IEL-depleted cell fractions. It is noteworthy that there was no evidence for further increases in interleukin-1β in any tissue later during the infection but that TNF mRNA levels rose steadily in the host spleen.

Unmasking of intestinal epithelial lateral membrane β1 integrin consequent to transepithelial neutrophil migration in vitro facilitates inv-mediated invasion by Yersinia pseudotuberculosis
-
McCormick BA, Nusrat A, Parkos CA, D'Andrea L, Hofman PM, Carnes D, Liang TW, Madara JL. Unmasking of intestinal epithelial lateral membrane β1 integrin consequent to transepithelial neutrophil migration in vitro facilitates inv-mediated invasion by Yersinia pseudotuberculosis. Infect Immun 1997, 65:1414-1421. A refreshing perspective with novel insights into the interactions between neutrophils and colon epithelial cells showing an effect of the interaction on the epithelium. The integrins that become exposed because of the neutrophil migration become attachment sites for Yersinia pseudotuberculosis, which can then infect the monolayer. This mechanism may help understand the chronic nature of inflammatory conditions typified by high numbers of neutrophils in the gut.

Cellular detachment and deformation induce IL-8 gene expression in human bronchial epithelial cells
-
Shibata Y, Nakamura H, Kato S, Tomoike H: Cellular detachment and deformation induce IL-8 gene expression in human bronchial epithelial cells. J Immunol 1996, 156:772-777. This paper is concerned with epithelial cells but not from the intestine. It shows that detaching bronchial epithelial cells by a number of means stimulates interleukin-8 production. Deformation following detachment is implicated in the stimulation by controlling cell shape with drugs.

Evidence for altered regulation of IkappaBα degradation in human colonic epithelial cells
-
Jobin C, Haskill S, Mayer L, Panja A, Sartor RB: Evidence for altered regulation of IkappaBα degradation in human colonic epithelial cells. J Immunol 1997, 158:226-234. The authors describe the unconventional pattern of IκBα degradation in response to interleukin-1 treatment, in human IEC lines and freshly isolated cells. The pattern of IκBα decay in Caco-2 cells resembled decay in monocytes and differed from all other IECs examined. Less NFκB translocated into the nucleus of IEC, which showed delayed IκBα degradation, but this did not affect interleukin-8 gene expression or protein levels, nor interleukin-1κ gene expression.
