•], characterizes the phosphorylation of the retinoblastoma protein (pRb) by different cyclin-dependent kinase (CDK)-cyclin complexes. Although the site of inactivation of pRb by CDK4-cyclin D1 is distinct in each paper (as is the functional assay used to used to test the site), these studies reach the same conclusions concerning differential pRb phosphorylation by CDK4-cyclin D1 and CDK2-cyclin complexes. In this paper, the authors show that only CDK4-cyclin D1 is able to functionally inactivate pRb as determined by a microinjection-based cell cycle assay. The phosphorylation site required for this effect was mapped to serine 795.

In this study, ectopic expression of cyclin E is shown to overcome the inhibitory effects of p16, a phosphorylation-deficient pRb and a dominant-negative DP1, thereby promoting S-phase entry. Hence, the most important functions of the E2Fs (proteins that are normally inhibited by the pocket proteins mentioned above) in cycling cells may be to turn on cyclin E, which on its own can induce cells to enter S phase.
