Fetcho Imaging neuronal activity during zebrafish behavior with a genetically encoded calcium indicator J Neurophysiol 90 2003 3986 3997 This study illustrates the use of cameleon, the genetically encoded ratiometric calcium indicator, to monitor neuronal activity during motor behaviors in zebrafish. It is the first study to report stable transgenic lines with a functional genetically encoded indicator in a vertebrate. Similar lines with other indicators have recently been produced in mice and these indicator lines will soon become as numerous as those expressing GFP.

Konnerth In vivo two-photon calcium imaging of neuronal networks Proc Natl Acad Sci USA 100 2003 7319 7324 This crucial technical innovation offers a way of labeling neurons in mammals and other species with calcium indicators in vivo, without disruption of their processes. The work enables the imaging of patterns of activity with single cell resolution in the brain of intact mammals by using an AM bulk-loading method in combination with multiphoton imaging. This, along with genetic labeling methods, promises to provide unprecedented views of the links between patterns of neural activity and behavior in mammals.

Although this work is in cultured cells, the non-linear approach should allow for deeper imaging in intact animals. This paper moves the field closer to the ultimate physiological goal of directly monitoring membrane potential at different parts of a neuron or many neurons in vivo, during behavior.

Beattie Mutations in deadly seven/notch1a reveal developmental plasticity in the escape response circuit J Neurosci 23 2003 8159 8166 This study shows that a genetic perturbation in a mutant line leads to the generation of extra cells that are seemlessly incorporated into functional circuits. It reveals how the nervous system can adapt during development to changes in the number of neurons within a network. This helps to explain how a behavior initiated by one cell in a fish can evolve into a behavior produced by many cells without compromising function in the process.

Fetcho Engrailed-1 expression marks a primitive class of inhibitory spinal interneuron J Neurosci 24 2004 5827 5839 This work reveals that the transcription code in spinal cord defines primitive functional classes of neurons. The study combines transgenic labeling with in vivo imaging and electrophysiological studies in a fashion that will soon become common-place in studies of the nervous system. It also shows how transcription factors can serve to define the relationships among cell types across species and reveal aspects of the evolution of circuits.

Goulding Genetic identification of spinal interneurons that coordinate left-right locomotor activity necessary for walking movements Neuron 42 2004 375 386 This study reports how a knockout of a transcription factor gene alters the projections of a class of neurons and in doing so changes the motor pattern in the limbs of mice. This suggests that the cells defined by a particular transcription factor are part of the CPG for limb movements. The study unites the transcription factor code used for spinal development with later circuit function and behavior.

Soffe Primitive roles for inhibitory interneurons in developing frog spinal cord J Neurosci 24 2004 5840 5848 This beautiful study explores the physiological roles of engrailed positive neurons in frog tadpoles and shows that this transcription factor marks a multifunctional cell type that might represent the evolutionary precursor of well-studied interneurons such as Renshaw cells in mammals. This work exemplifies how the essential functional roles of neurons and their evolutionary specializations can be uncovered through a combination of developmental and functional information.
