Blood was diluted at 1:1 with a PBS sterile solution to a total volume of 2 mL. The diluted blood was placed in 3 mL of Histopaque-1077 (Sigma-Aldrich Inc., Dorset, UK) in a 15-mL tube. The samples were centrifuged in an Eppendorf (Hamburg, Germany) centrifuge at 400 × g for 30 min at room temperature. The resulting buffy coat of white cells over the red blood cell layer was collected and transferred to a 15-mL tube. The cells were washed twice with 4 mL of PBS and centrifuged at 400 × g for 7 min. The resulting sediment was resuspended in 1 mL of a paraformaldehyde solution in PBS at 2% for cell fixation. Thirty min after resuspension, the cells were centrifuged at 400 × g for 7 min., the supernatant was discarded, the cells were washed twice with PBS and centrifuged at the same speed and for the same amount of time, and the final sediment was resuspended in PBS with 1% BSA. Cells were counted in a Neubauer chamber. Single staining was performed for all antibodies using 2 μL of the primary antibody diluted in 20 mL of PBS (pH 7.4). This diluted solution was then mixed with mononuclear cells and maintained at room temperature in the dark for 20 min. After this period of incubation, the cells were washed with 2 mL of PBS and centrifuged at 400 × g for 7 min. The supernatant was discarded. The cells received the secondary antibody (5 μL, 1:200 dilution) or streptavidin FITC (Invitrogen, Carlsbad, CA; diluted at 1 mL:10 μL PBS), as required. The cells were maintained at room temperature in the dark for 20 min and then washed with 2 mL of PBS and centrifuged. The resulting sediment was resuspended in 250 μL of PBS with BSA. All samples were submitted to cytometry within 2 hours of staining. Flow cytometry was performed using a flow cytometer (FACSCalibur, Becton Dickinson, Franklin Lakes, NJ). Green fluorescence (FITC fluorescence) was detected in the FL1 channel (nm 530/30), and orange fluorescence was detected in the FL2 channel (nm 585/42). At least 10,000 events were analysed in the lymphocyte gate (based on frontal and lateral dispersion). Data were analysed using the FlowJo (TreeStar Inc., Ashland, OR) software program.

To evaluate the T lymphocytes (CD3+ cells) in the intestinal mucosa, sections of the duodenum, jejunum, ileum, and cecum were deparaffinized in an oven at 60°C for 10 min in xylene pure and rehydrated through a series of graded alcohols (ethanol). Antigen was recovered using a citrate buffer with a pH of 6.0 in a water bath at 100°C for 20 min. Endogenous peroxidase blocking (solution containing hydrogen peroxide at 3%) and protein blocking were performed for 8 and 5 min, respectively. Anti-CD3 was used as the primary antibody (Polyclonal rabbit anti-human CD3, DAK O, Glostrup, Denmark) and diluted (Dako Antibody diluent, DAK O) at 1:300 for 40 min. The reaction was detected by antirabbit and antimice secondary antibodies using the same amplification system (Advance kit, DAK O,) for 40 min. and a chromogen (DAB kit, Abcam, Cambridge, MA) for staining for 5 min. Counterstaining was performed with hematoxylin for 5 min after dehydration and preparation of the slides.
