This study analyses the regulation of the expression of the different variants of the BK channel during the development of alcohol tolerance by miR-9. It highlights the importance of the careful analysis of the different splice variants that are expressed in a tissue-specific manner or upon specific conditions, as they can be submitted to distinct regulation processes. This study provides, to our knowledge, the first demonstration of ion transport regulation by miRs using both human samples and rat experimental models of myocardial infarction. Not only do the authors show that miR-1 expression is modulated in the heart of patients, they also directly prove that miR-1 overexpression is responsible for arrhythmia by transfering miR-1 or its inhibitor in vivo in the rat heart and studying the effects of this overexpression by electrocardiograms and patch-clamp recordings. This study describes the regulation of the expression of the serine-threonine kinase WNK1, an important regulator of ion transport, by miR-1 92 in vitro and also provides evidence that this microRNA could be involved in the regulation of the kinase expression by aldosterone in the kidney. In addition, this study was the first to show that the expression of microRNAs can be regulated by physiological stimuli, such as sodium depletion or potassium load, in the kidney.
