It was initially believed that inhibition of MMP-1 or MMP-14 was the cause of MSS. Studies on MMP-14 KO mice support MMP-14 being a strong candidate for MSS: see Ref. 4b. However, clinical data with CP-544439, a non-selective hydroxamic acid-based MMP-13 inhibitor showed MSS side effects even though it is inactive against MMP-1. See:
