Mapping methyl jasmonate-mediated transcriptional reprogramming of metabolism and cell cycle progression in cultured Arabidopsis cells
-
The authors monitored the impact of methyl-jasmonate in cell cycle progression of cultured Arabidopsis cells and described two independent waves of responses. The first wave corresponds to the induction of genes related to JA biosynthesis or transcriptional regulation, including MYC2 and JAZ. The second transcriptional wave leads to a repression of cell cycle in G2 and to the activation of phenylpropanoid metabolism. Moreover, direct involvement of transcription factors in the regulation of JA biosynthesis was demonstrated by their binding to LOX3 promoter or by transient transactivation assays.

COI1 is a critical component of a receptor for jasmonate and the bacterial virulence factor coronatine

The authors demonstrate that a functional COI1 protein is required for JA perception in tomato. Coronatine (COR) and JA-Ile can promote the interaction between tomato COI1 and JAZ proteins (SlCOI1 and SlJAZ1/3) in PD experiments in vitro. Using radio-labelled COR authors demonstrate that this molecule can be pulled-down with the COI1-JAZ complexes, and that JA-Ile can compete with COR binding, indicating that both molecules are recognised by the same receptor. Binding of COR to the COI1-JAZ complex requires COI1 and is greatly impaired by a point mutation in the putative ligand-binding pocket of COI1, suggesting that COI1 may be the COR and JA-Ile receptor. Authors finally show that the conserved Jas motif of SlJAZ3 is necessary and sufficient for hormone-induced COI1-JAZ3 interaction.

The JAZ family of repressors is the missing link in jasmonate signalling
-
The characterisation of jai3-1, a dominant JA-insensitive mutant, allowed the identification of JAI3/JAZ3 and the related JAZ family of repressors in Arabidopsis. jai3-1 encodes a mutant protein that lacks the C-terminal Jas motif. Constitutive expression of a similar truncated form of JAZ3 lacking the Jas motif (JAZ3ΔJas) also renders JA-insensitive plants.

COI1 for proteasome degradation in response to jasmonate, the JAZ3ΔJas mutant protein is resistant to degradation and impairs JA-dependent instability of several JAZ proteins. Moreover, JAZs and MYC2 are involved in a negative regulatory feed-back loop that provides a mechanistic explanation for the pulsed response to jasmonate and the subsequent desensitisation of the cell.

A critical role of two positively charged amino acids in the Jas motif of Arabidopsis JAZ proteins in mediating coronatine- and jasmonoyl isoleucine-dependent interactions with the COI1 F-box protein
-
By means of yeast two-hybrid assays, the authors show that JA-Ile and its structural analogue COR can promote interaction between Arabidopsis COI1 and several JAZ proteins. Confirming previous results [19] the Jas motif of JAZ proteins is the only domain required for hormone-induced COI1-JAZ interactions. Furthermore two positively charged amino acid residues within this domain are essential for coronatine-dependent COI1-JAZ interactions but do not affect JAZ1/9-MYC2 interactions. Transgenic plants expressing JAZ1 carrying mutations in both residues exhibited JA-insensitive phenotypes showing the relevance of these two amino acids of the Jas motif in COI1-JAZ1 interactions in vivo.

The conserved TIFY motif within the ZIM domain of JAZs mediates homomeric and heteromeric interactions between most members of the Arabidopsis JAZ family. A thorough yeast two-hybrid analysis is done to check JAZ-JAZ interactions. In wounded leaves authors also find the natural occurrence of a novel splice variant (JAZ10.4) of JAZ10 that lacks the entire Jas motif. JAZ10.4 has the ability to interact with MYC2 but is resistant to JA-induced degradation since it is unable to interact with COI1 and its overexpression confers severe JA-insensitivity. Dominant JA-insensitive phenotype resulting from JAZ10.4 overexpression is suppressed by mutations in the TIFY domain that obstruct JAZ10.4-JAZ interactions. A model is proposed in which JAZ10.4-containing protein complex directly impairs the activity of JA-responsive transcription factors.

Identification of (+)-7-iso-JA-L-Ile as the endogenous bioactive jasmonate in Arabidopsis. (-)-JA-l-Ile and (+)-7-iso-JA-l-Ile diastereomers are in equilibrium in the cell and in chemically synthesised samples (ratio 94:6), the latter being structurally more similar to coronatine. Purification of these compounds allowed testing of their promotion of COI1-JAZ interaction separately in PD assays. These experiments demonstrated that pure (-)-JA-l-Ile is inactive and that (+)-7-iso-JA-l-Ile is the active form of the hormone. In addition, pH changes in vitro promote conversion of (+)-7-iso-JA-l-Ile to the inactive (-)-JA-l-Ile. (+)-7-iso-JA-l-Ile is stable at cellular pH and therefore its conversion to (-)-JA-l-Ile in the cell suggests that epimerisation is a regulatory mechanism of hormone activity. Authors also propose methyl-esterification as an additional mechanism of regulation of hormone activity.
