Mapping methyl jasmonate-mediated transcriptional reprogramming of metabolism and cell cycle progression in cultured Arabidopsis cells

The authors monitored the impact of methyl-jasmonate in cell-cycle progression of cultured Arabidopsis cells and described two independent waves of responses. The first wave corresponds to the induction of genes related to JA biosynthesis or transcriptional regulation, including MYC2 and JAZ. The second transcriptional wave leads to a repression of cell cycle in G2 and to the activation of phenylpropanoid metabolism. Moreover, direct involvement of transcription factors in the regulation of JA biosynthesis was demonstrated by their binding to LOX3 promoter or by transient transactivation assays.

MYC2 differentially modulates diverse jasmonate-dependent functions in Arabidopsis
-
By means of transcriptomic profiling and subsequent phenotypic and biochemical analysis, the authors provided an in-depth characterization of MYC2 functions as coordinator of JA-induced responses. Consistent with a previous hypothesis [27] authors showed that MYC2 positively regulates tolerance to oxidative stress, flavonoid biosynthesis, and resistance to insect herbivory and negatively regulates Trp metabolism, leading to JA-dependent synthesis of defensive compounds such as indole glucosinolates. It was suggested that the dual modulation of JA responses exerted by MYC2 could be because of its capacity to regulate intermediate transcription factors. Authors also determined the sequence specificity of MYC2 binding to DNA in vitro. Finally, they found that MYC2 negatively regulates its own expression and this mechanism is pointed out as a fine-tuning of JA signaling.

The application of JA-Ile to plants impaired on JA biosynthesis (lox3 mutant) did not fully restore their resistance levels, meaning that other oxylipins should complement the activity of JA-Ile in defense. Hormone transport experiments suggested that JA-Ile is not a long-distance signaling molecule instead being synthesized in locus upon arrival of a mobile signal.
