Highly efficient synthesis of β-amino esters via Mannich-type reaction under solvent-free conditions using ZnCl2 catalyst

    • The synthesis of aspartic acid derivativate 4: a solution of compound 3a (29 mg, 0.85 mmol, sodium chloride (16 mg, 2.8 mmol) and water (61 mg, 3.4 mmol) in DMSO (10 mL) was heated at 100 °C for 4 h. The reaction was cooled to ambient temperature before being diluted with ethyl acetate (50 mL, The organic phase was washed with water and brine before being dried over MgSO4 and concentrated in vacuo. The crude product was purified by flash chromatography (ethyl acetate:light petroleum, 1:6) to yield the desired product 4 as a yellow liquid (65, 1H NMR (CDCl3, 300 MHz, δ ppm, 1.23-1.28 (t, 3H, J, 7.2 Hz, 2.84-2.86 (d, 2H, J, 6.0 Hz, 3.71(s, 3H, 3.74 (s, 3H, 4.16-4.23 (m, 3H, 6.64-6.67 (d, 2H, J, 9.0 Hz, 6.76-6.78 (d, 2H, J, 9.0 Hz, 13C NMR (CDCl3, 75 MHz, δ ppm, 14.51, 37.81, 52.30, 55.30, 56.01, 61.85, 115.08, 116.00, 141.58, 153.37, 171.18, 172.65; MS (ESI) m/z 282, M+1
