22. TM transfection reagent, with reporter (pGal5-TK-pGl3) and expression plasmids (pGal4-hRARα, γor β). Cells were incubated at 37 °C. After 24 h, cells were trypsinised and seeded in 96-well plates and incubated for 6 h in DMEM containing 0.2% FCS. Cells were then incubated 16h in DMEM 0.2% FCS and increasing concentrations of the compounds tested or vehicle (DMSO). At the end of the experiments, cells were washed once with PBS and the luciferase and the β-galactosidase assays were performed.
