Important conclusions drawn from this study include the identification of Eya1, Six1 and Six4 as definitive markers of the pan-placodal domain and the determination of the location of this domain, with respect to the neural plate and neural crest forming regions. Finally this study defines the earliest stages at which each placode appears as a molecularly distinct unit.

Whereas misexpression of various transcription factors (both in this paper and others) results in the ectopic expression of δ-crystallin in the region adjacent to the lens, none has shown expansion of the δ-crystallin positive region to the entire head ectoderm. Here, the authors show that co-expression of L-Maf and Sox2 not only induces δ-crystallin strongly in the entire head ectoderm, but also imparts lens fiber-like morphology to these cells.

Multiple enhancers governing different spatiotemporal aspects of Sox2 expression were identified using electroporation of the early chick embryo. Enhancers include those specific for various placodes - lens, nasal and otic - as well as various neural enhancers.
